First of all, this is still an interview. It shouldn't be a formal conversation (nor should most interviews be very formal), but you should still do your research, practice some answers, and prepare your own questions. In addition, you'll want to express your enthusiasm for the opportunity and talk to them even if you're not very interested in the position (you can always turn it down later). Otherwise, let the call go to voicemail and call the interviewer again as soon as possible (when you're ready)).
